چکیده
In article number 2000985, Marek Piliarik and co-workers develop a new light microscopy technique for label-free detection localization of conformation changes subsequent depolymerization tubulin segments from tip disassembling microtubule. High-speed imaging the transient structural fluctuations suggests stochastic mechanism microtubule dynamic instability.

Dynamics and organization of cortical microtubules as revealed by superresolution structured illumination microscopy.
Plants employ acentrosomal mechanisms to organize cortical microtubule arrays essential for cell growth and differentiation. Using structured illumination microscopy (SIM) adopted for the optimal documentation of Arabidopsis (Arabidopsis thaliana) hypocotyl epidermal cells, dynamic cortical microtubules labeled with green fluorescent protein fused to the microtubule-binding domain of the mammal...
متن کاملNanoscopic lipid domain dynamics revealed by atomic force microscopy.
Intrinsic heterogeneities, represented as domain formations in biological membranes, are important to both the structure and function of the membranes. We observed domain formations in mixed lipid bilayers of dipalmitoylphosphatidylcholine (DPPC), dilauroylphosphatidylcholine (DLPC), and cholesterol (chol) in a fluid environment using an atomic force microscope (AFM). At room temperature, we de...
متن کاملStructural organization of nuclear lamins A, C, B1, and B2 revealed by superresolution microscopy
The nuclear lamina is a key structural element of the metazoan nucleus. However, the structural organization of the major proteins composing the lamina is poorly defined. Using three-dimensional structured illumination microscopy and computational image analysis, we characterized the supramolecular structures of lamin A, C, B1, and B2 in mouse embryo fibroblast nuclei. Each isoform forms a dist...
متن کاملSpatial separation of Xist RNA and polycomb proteins revealed by superresolution microscopy.
In female mammals, one of the two X chromosomes is transcriptionally silenced to equalize X-linked gene dosage relative to XY males, a process termed X chromosome inactivation. Mechanistically, this is thought to occur via directed recruitment of chromatin modifying factors by the master regulator, X-inactive specific transcript (Xist) RNA, which localizes in cis along the entire length of the ...
متن کاملHeterogeneous architecture of vertebrate kinetochores revealed by three-dimensional superresolution fluorescence microscopy
The kinetochore is often depicted as having a disk-like architecture in which the outer layer of proteins, which engage microtubules and control checkpoint signaling, are built on a static inner layer directly linked to CENP-A chromatin. Here, applying three-dimensional (3D) structural illumination microscopy (SIM) and stochastic optical reconstruction microscopy (STORM) to Xenopus egg extracts...
